Problem-Solving Test: Telomere Replication

The Nobel Prize in Physiology or Medicine in 2009 was awarded to Elizabeth H. Blackburn, Carol W. Greider, and Jack W. Szostak for the discovery of "how chromosomes are protected by telomeres and the enzyme telomerase." The discovery has important implications in the processes of cellular aging and carcinogenesis. Telomeres are satellite DNA regions at the tips of eukaryotic chromatids. In human cells they consist of thousands of tandemly repeated hexanucleotides. This article presents a problem-solving test on telomere replication, which consists of three parts. First, the difficulties of replicating the ends of a linear DNA molecule are exposed; solving the multiple-choice questions (MCQs) related to this problem requires one's pre-existing knowledge of the mechanism of DNA replication. The second part of the test deals with the features of telomerase. Finally, the results of an experiment to analyze the role of telomerase are presented. (Contains 4 figures.)
