Described is a laboratory exercise designed to introduce students to the theory and practice of gene mapping including RNA extraction, sucrose density gradient centrifugation, labelling of nucleic acids in vitro, DNA extraction, digestion of DNA with restriction enzymes, and the southern hybridization analysis.
