Illustrating the Effect of pH on Enzyme Activity Using Gibbs Energy Profiles

Gibbs energy profiles provide students with a visual representation of the energy changes that occur during enzyme catalysis, making such profiles useful as teaching and learning tools. Traditional kinetic topics, such as the effect of pH on enzyme activity, are often not discussed in terms of Gibbs energy profiles. Herein, the symbolism of Gibbs energy profiles is utilized to derive expressions for the changes in Gibbs energy associated with the apparent enzyme kinetic parameters K[subscript S][superscript app], k[subscript cat][superscript app], and (k[subscript cat]/K[subscript M])[superscript app] and to describe their behavior as a function of pH under rapid equilibrium conditions. This approach provides students with an alternative way of understanding the effect of pH on enzyme kinetics that is complementary to the traditional derivations using pH-dependent kinetic equations.
